ABOUT IGF-1 LR3: Insulin-like growth factor long arginine 3 (IGF-1 LR3) is a modified version of insulin-like growth factor-1 (IGF-1). It contains the complete human IGF-I protein sequence with a substitution of arginine for glutamic acid at the third position and also has additional 13 amino acids at its N-terminus making it a total of 83 amino acids compared to the 70 of IGF-1.
The consequences of these modifications are that IGF-1 LR3 retains the pharmacological activity of IGF-1 as an agonist of the IGF-1 receptor, with improved metabolic stability and a very low affinity for the insulin-like growth factor-binding proteins (IGFBPs). This makes it three times more potent than IGF-1, with a significantly longer half-life of about 20–30 hours (relative to IGF-1’s half-life of about 12-15 hours).
IGF-1 LR3 is a hormonal mediator of growing stature. Under normal circumstances, GH (growth hormone) binds to its receptor in the liver, and other tissues, and stimulates the synthesis/secretion of IGF-1. In target tissues, the Type 1 IGF receptor, which is homologous to the insulin receptor, is activated by IGF-1 LR3, leading to intracellular signaling, which stimulates multiple processes leading to stature growth.
