What is Thymosin Beta 4

Thymosin Beta 4 is a natural protein in humans encoded by the TMSB4X gene. It is also known as Timbetasin as published by the World Health Organization. It is made up of 43 amino acids with molecular weight of 4,921 g/mol.

Thymosin Beta 4 is water soluble, regenerative, and is found all tissues and cell types except in the red blood cells. However, it can be found in other body fluids such as tears, saliva, wound fluids, and the cerebrospinal fluid.

What is TB-500

TB-500 as it is commonly known is actually a veterinary preparation used for race horses to prevent adhesions. TB-500 is a synthetic version of Thymosin Beta 4. Its main component is the peptide segment (17)LKKTETQ(23), which is also the region in Thymosin Beta 4 responsible for binding with actin and exhibiting effects such as angiogenesis, wound healing, and cell migration.

Effects of Thymosin Beta 4

Thymosin Beta 4 is the major G-actin-sequestering peptide in mammals, several published studies describes its biological effects. This includes promotion of angiogenesis or blood vessel formation, cell migration, cell survival, stem cell differentiation, and modulation of cytokines and chemokines. 

Effects of TB-500

Studies involving TB-500 had a wide range of result regarding its effect. This includes promoting angiogenesis, endothelial cell differentiation, collagen deposition, keratinocyte migration, and anti-inflammatory effects.

Technically, TB-500 has the same effects as that of Thymosin Beta 4 since the former is just the isolated active region of the latter.

Actin-binding. Thymosin Beta 4/TB-500 forms a complex with Globular actin. These globular actin monomers polymerizes with each other Filamentous actin or F-actin. These actins together with other actin-binding proteins comprise the cellular microfilaments.

The release of globular actin monomers from Thymosin Beta 4/TB-500 is important in tissue formation, wound healing, and immune response.

Tissue Regeneration. Animal experiment and tissue culture studies have shown that Thymosin Beta 4/TB-500 promotes cell migration, re-epithelialization, and angiogenesis or blood vessel formation. These effects are important in tissue repair and regeneration.

Studies conducted in mice models has also shown Thymosin Beta 4/TB-500 to help recover from autoimmune encephalomyelitis, which is an animal model equivalent to multiple sclerosis.

Wound Healing. As mentioned above, Thymosin Beta 4/TB-500 has the ability to promote cell migration and re-epithelialization. These activities has shown to greatly help in healing dermal and corneal wounds conducted in animal studies. It also has antimicrobial property that helps prevent microbial infection at wound sites that could further damage the tissue.

It has been used as topical treatment in rats and mice models where it successfully treated heptanol debridement and alkali injury in the skin and the eyes. 

It is also currently being investigated for its possible use in helping repair wounds in the heart caused by heart attack and heart disease. A study conducted on rats has shown that it stimulates formation of new heart muscle cells.

Anti-Inflammatory. Thymosin Beta 4/TB-500 regulates inflammatory chemokines and cytokines response. Chemokines and cytokines are molecular messengers that tells white blood cells to migrate at the site of wound thus causing inflammation, swelling, and possible further tissue damage.

The downregulating effect of Thymosin Beta 4/TB-500 has on chemokines and cytokines decreases inflammatory response. It also appears to be blocking TNF-alpha cytokine release, reduces matrix metalloproteinases (MMPs), and activates nF kappa-B – all of which contributes to decreased inflammatory response.

Anti-Apoptosis. Apoptosis or cell death is normal for most cells, but premature apoptosis can cause a lot of problems. Thymosin Beta 4/TB-500 has shown to prevent cell death caused by infections, toxins, or lack of blood supply, thus promoting cell survival.

            It is also cardioprotective and has shown neuroprotective activity in both in vivo and in invitroanimal studies. Thymosin Beta 4/TB-500 given via intracerebroventricular administration has been found to protect the death of neurons in the hippocampal region even after kainic acid treatment.
